Surfactant Stabilizer (Polysorbate)

A surfactant stabiliser is a non-ionic excipient, usually polysorbate 20 or 80, added to occupy interfaces so that the peptide is not the molecule that unfolds at them.

A surfactant stabiliser is an excipient added not for bulk solubility but to win a competition for surfaces. Polysorbate 20 and polysorbate 80, and to a lesser extent poloxamer 188, are amphiphilic molecules that reach air-liquid and liquid-solid interfaces far faster than a peptide can, so the adsorbed film that forms is surfactant rather than partly unfolded drug. They also bind weakly to exposed hydrophobic patches on the molecule. Concentrations are very low, in the hundredths of a percent, because the effect is interfacial rather than bulk.

This is why a polysorbate appears in the excipient list of most peptide and biologic injections, and why it protects against agitation and container surfaces and against nothing else. Polysorbates are themselves unstable: they hydrolyse and oxidise over shelf life, releasing free fatty acids that can precipitate as visible particles late in a product's life, and residual peroxides in the raw material can oxidise methionine and tryptophan residues in the drug, which is why peroxide specifications exist.

The tradeoff is unavoidable rather than clever. Removing the surfactant exposes the peptide to interfacial damage; keeping it introduces a degradation pathway and a particle source that must then be controlled. Choosing between polysorbate 20 and 80 is largely a question of which hydrolysis and oxidation profile is less damaging to that particular molecule.

The recurring error is expecting the wrong kind of protection. A surfactant does nothing about deamidation, thermal degradation or oxidation of the peptide by other routes, so a formulation containing one is not thereby stable. The other misreading treats a surfactant-free grey-market product as purer or more natural, when the absence of a stabiliser usually means no formulation development happened at all.
