Silicone Oil

Silicone oil is the polydimethylsiloxane lubricant applied to syringe barrels and stoppers so the plunger moves smoothly, and it is also a common source of droplets in the drug solution.

Silicone oil in a parenteral context means polydimethylsiloxane applied as a thin layer inside a syringe barrel and onto elastomeric stoppers. Without it the plunger cannot break loose and glide at a force a hand can apply, so it is not optional. It is applied either as a sprayed emulsion, leaving a mobile film, or baked on and cross-linked to the glass, which is far more durable and sheds much less. Because the film is mobile and the drug rests against it for the whole shelf life, some silicone migrates into solution as microscopic droplets.

Those droplets have a specific analytical consequence. Light obscuration, the compendial method for subvisible particles, counts anything that blocks light, so a silicone droplet and a protein aggregate register identically. Separating them needs an imaging method such as micro-flow imaging, where droplets appear as optically distinct spheres and aggregates are irregular. Silicone-free polymer syringes exist for sensitive products, and polysorbate in the formulation can strip silicone from the barrel wall over time.

The oil-water interface is also a nucleation site, so silicone is more than an analytical nuisance: adsorption at droplet surfaces can seed genuine protein aggregation, particularly under agitation. Formulation choice trades lubricity against particle burden, and a rising particle count in a stability study is uninterpretable until the two populations are told apart.

The error worth naming is reading a particle count as protein aggregation when the particles are silicone, or dismissing a real aggregate signal as probably silicone. Both happen. The visible version appears after repeated intravitreal injections, where silicone droplets present as floaters that patients and clinicians can mistake for a disease process rather than a container-closure artefact.
