Diluent
A diluent is the liquid used to dissolve a lyophilised powder or dilute a concentrate, and it is a formulation component whose composition and pH can determine whether the product survives.
A diluent is the fluid added to a dry or concentrated drug to bring it to a usable solution. Common diluents are water for injection, sodium chloride solution, dextrose solution and manufacturer-supplied proprietary vehicles that may contain a buffer, a surfactant or a preservative. Which one is specified is settled during development by compatibility testing, because the diluent contributes the final pH, ionic strength and tonicity, and any of those can decide whether the peptide stays in solution.
Several protein products are co-packaged with their own diluent for that reason. Somatropin has historically been supplied with preserved vehicles containing metacresol or benzyl alcohol in some formats and a preservative-free vehicle in others, so a neonatal presentation avoids the preservative entirely. Where labelling names a specific diluent, that is a compatibility finding rather than a commercial preference, and substituting another vehicle leaves the data that supported approval.
The diluent sets two separate risks at once. Its composition governs chemical compatibility, since an unbuffered vehicle can let a solution drift toward the peptide's isoelectric point and precipitate, while a saline vehicle raises ionic strength in a way some peptides tolerate poorly. Independently, the preservative governs how long the solution can reasonably be kept, a microbiological question rather than a chemical one.
The frequent error is treating diluents as interchangeable water. Dextrose solutions are mildly acidic and reactive toward some molecules, saline is not neutral for a salt-sensitive peptide, and a preserved vehicle is unsuitable where labelling forbids the preservative. Grey-market usage stretches the term to cover any clear liquid, which is how non-pharmaceutical solvents end up in preparations described as reconstituted.
