Crude vs Purified Peptide
Crude peptide is the unpurified mixture recovered straight from the resin, while purified peptide is the fraction collected after preparative chromatography has discarded most of that mass.
Crude peptide is everything that precipitates after cleavage: the full-length target together with deletion sequences, truncated chains, epimers, incompletely deprotected and alkylated species, residual salts and the counterion from the cleavage acid. Purified peptide is the narrow fraction collected when that mixture is run through preparative chromatography and the peak of interest is cut, pooled and lyophilised. The two words describe positions in one process, not two grades of the same substance.
The gap between them grows with length. A routine short peptide might come off the resin at sixty to eighty percent area purity; a long sequence with aggregating stretches can be well under half. Purification then trades mass for purity, and recovering only a modest fraction of the crude as high-purity product is entirely normal. This is why purification rather than synthesis dominates the cost of peptide manufacture, and why the price gap between crude and high-purity material is not a margin decision.
Whether crude is acceptable depends on what the impurities do. For screening libraries, epitope mapping or a qualitative binding experiment, a mixture is often fine. For anything where the impurity fraction has its own pharmacology it is not, because a deletion sequence and an epimer are different molecules with their own receptor behaviour and immunogenic potential. That fraction cannot be assumed inert because it is chemically related to the target.
The commercial version is a listing pairing a very low price with a very high stated purity, two claims difficult to hold together given what purification costs. The stated figure is also easy to flatter: a short gradient that never elutes late-running impurities, detection at a wavelength where non-aromatic species barely absorb, or an integration treating a raised baseline as baseline.
